Introduction

The chemical name of fludarabin is 9-1 β D-arabifurose-2-fluoroadenosine is an anti-tumor drug. Fludarabin is an anti metabolic fluoropurine nucleoside analogue with high selectivity towards lymphocytes. It can inhibit the repair of DNA in quiescent Chemicalbook cells and the synthesis of DNA in dividing cells. It also promotes apoptosis in these cells and has shown good therapeutic effects. Currently, it is widely used in the treatment of various hematological diseases, especially chronic B-lymphocytic leukemia.

Function

This drug is a fluorinated nucleotide analogue of adenosine arabinose, which can relatively resist the deamination effect of adenosine deaminase. Fludarabin phosphate is rapidly dephosphorylated to form 2F ara-A, which can be taken up by cells and then phosphorylated by intracellular deoxycytidine kinase to form the active triphosphate chemicalbook salt 2F ara-ATP. This metabolite can be produced by inhibiting nucleotide reductase and DNA polymerase( α,δ and ε), DNA primers and DNA ligases inhibit DNA synthesis. In addition, it can partially inhibit RNA polymerase II, thereby reducing protein synthesis.

Application

Its phosphate group is rapidly cleaved in the bloodstream, and the remaining 2-fluoroadenosine (Fara-A) enters the cells and is phosphorylated into the active metabolite Farar ATP, exerting its effects. Used for chronic lymphocytic leukemia.
